Background
TOP2A (DNA topoisomerase II alpha) is an enzyme that controls the topological states of DNA through ATP-dependent breakage, passage, and rejoining of double-stranded DNA. It is specifically inhibited by the intercalating agent amsacrine. Both eukaryotic topoisomerase I and II can relax negative and positive supercoils, whereas prokaryotic enzymes relax only negative supercoils. Phosphorylation has no effect on catalytic activity. TOP2A generally localizes in the nucleoplasm and functions as a homodimer interacting with COPS5.
