The Butyrylcholinesterase Fluorescent Activity Kit is a research-use-only fluorescent assay designed for the quantification and detection of butyrylcholinesterase (BChE) activity in serum and plasma samples.

A fluorescence microplate reader capable of detecting emission at 510 nm with excitation at 390 nm is required.

Background
Butyrylcholinesterase (BChE) belongs to the same structural class of proteins as acetylcholinesterase (AChE). The 440 kDa tetrameric glycoprotein is found in blood, kidneys, intestine, liver, lung, heart, and the central nervous system.
Many species such as human, horse, and mouse exhibit high BChE activity in plasma, whereas rats have higher acetylcholinesterase activity.
BChE preferentially acts on butyrylcholine but also hydrolyzes acetylcholine.
This assay has been validated for serum, EDTA, and heparin plasma from various species.
Assay Principle
The kit quantitatively measures BChE activity using a human BChE standard to generate a standard curve.
A proprietary non-fluorescent reagent reacts with the thiol product of the enzymatic reaction between BChE substrate and BChE, forming a fluorescent product measurable at 510 nm (emission) and 390 nm (excitation).

BChE Unit Definition
One unit hydrolyzes 1.0 μmol of butyrylcholine to choline and butyrate per minute at pH 8.0 and 37°C.
