Target Information

FAS (Fas, CD95, APO-1) is a 46 kDa transmembrane glycoprotein functioning as a cell death receptor in the TNFR superfamily. It also exists in a soluble 26 kDa form. Activation of FAS triggers aggregation of FADD domains, forming the death-inducing signaling complex (DISC) and activating caspases, leading to apoptosis. FAS can also activate NF-kappaB, MAPK3/ERK1, and MAPK8/JNK, contributing to proliferative signaling in fibroblasts and T cells. Multiple alternatively spliced variants exist, some lacking the transmembrane domain, which may regulate apoptosis negatively. The Fas/Fas ligand pathway is implicated in diseases such as AIDS, hepatitis, and cancer, and plays a role in antiviral immune responses.
