The antibody is intended for professional laboratory use to detect insulin in formalin-fixed, paraffin-embedded tissue sections using manual qualitative immunohistochemistry (IHC). It should be used after the primary diagnosis of tumor by conventional histopathology.

The antibody recognizes insulin, a 51-amino acid polypeptide composed of A and B chains connected through C-peptide. Proinsulin is cleaved into insulin and the C-terminal residue. Insulin regulates glucose, amino acid, and ion transport, and promotes glycogen, triglyceride, and protein synthesis. Insulin deficiency leads to diabetes mellitus. The main storage site is the pancreatic islets. Antibodies to insulin are important markers for beta-cell and insulinoma studies.

Pretreatment of deparaffinized tissue with heat-induced or enzymatic epitope retrieval is recommended. IHC staining allows visualization of antigens through sequential antibody applications and chromogenic substrate reaction, observed under a light microscope for diagnostic interpretation.

A positive tissue control must accompany each staining procedure. This control should include both positive and negative staining components. External positive control tissues should be freshly prepared, fixed, and embedded similarly to patient samples. Weakly positive tissues are preferred to detect subtle changes in antibody sensitivity or reagent stability.

Negative controls (internal or external) should show absence of specific staining to verify assay specificity. If specific staining occurs in negative controls, results must be considered invalid.

Target Information

Insulin is a peptide hormone secreted by pancreatic beta cells, regulating carbohydrate, protein, and lipid metabolism. It promotes glucose uptake, glycogen storage, and protein synthesis. Insulin is derived from proinsulin, which is cleaved into A and B chains linked by disulfide bonds and one C-peptide molecule. Insulin deficiency causes diabetes mellitus. It is also expressed in tumors of beta-cell origin such as insulinoma.
