Target Information
FAM21C는 WASH core complex의 구성 요소로서, endosome 표면에서 Arp2/3 complex를 활성화하여 actin polymerization을 유도합니다. 이는 endosome sorting 과정에서 수송 중간체로 작용하는 tubule의 절단에 중요한 역할을 합니다. 또한 phospholipids 및 retromer CSC의 VPS35와 결합하여 WASH core complex를 endosome 막으로 모집하며, F-actin-capping protein dimer의 결합을 매개하여 소포 절단에 필요한 국소적 F-actin 중합을 촉진합니다.
FAM21C는 endosome-to-plasma membrane trafficking 및 SNX27-retromer 의존적 cargo 단백질(예: GLUT1)의 재활용에도 관여하며, DNAJC13, ENTR1, ANKRD50 등의 retromer CSC 하위단위 VPS35 결합에 필요합니다. 또한 CCC 및 retriever complex의 COMMD1, CCDC93, VPS35L 등의 endosomal 모집에도 필수적입니다.
