Mammalian mitochondrial ribosomal proteins are encoded by nuclear genes and assist in protein synthesis within the mitochondrion. Mitochondrial ribosomes (mitoribosomes) consist of a small 28S subunit and a large 39S subunit, with approximately 75% protein and 25% rRNA composition, unlike prokaryotic ribosomes where this ratio is reversed. Prokaryotic ribosomes also contain a 5S rRNA, which mammalian mitoribosomes lack.

The MRPS12 gene encodes a 28S subunit protein belonging to the ribosomal protein S12P family. This protein is a key component of the ribosomal small subunit, controlling decoding fidelity and susceptibility to aminoglycoside antibiotics. The mitochondrial seryl-tRNA synthetase gene is located upstream and adjacent to MRPS12, and both are candidates for the autosomal dominant deafness gene (DFNA4). Splice variants differing in the 5' UTR have been found; all encode the same protein.
