Product Specific Information

The IM7 monoclonal antibody reacts with all isoforms of mouse CD44 (Pgp-1). CD44 is expressed by hematopoietic and non-hematopoietic cells. Bone marrow myeloid cells and memory T cells highly express this antigen, and peripheral B and T cells can upregulate CD44 expression. CD44 functions as an adhesion molecule through its binding to hyaluronate, an extracellular matrix component.

Applications Tested: Mouse splenocytes (≤ 0.125 µg/test)

A test is defined as the amount of antibody that stains a cell sample in a final volume of 100 µL. Cell numbers may range from 10⁵ to 10⁸ cells/test. Titration is recommended for optimal assay performance.

Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Use a 610/20 bandpass filter. Ensure the instrument supports this fluorochrome.

When using multiple Super Bright conjugates, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize polymer interactions.

Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.

Target Information

CD44 is a 100 kDa type 1 transmembrane glycoprotein expressed on human leukocytes, brain white matter, and epithelial cells. It binds hyaluronic acid (HA) and mediates cell-cell interactions, adhesion, and migration. CD44 plays roles in lymphocyte activation, homing, and may be linked to metastatic potential in carcinomas.

Multiple isoforms exist due to complex alternative splicing, contributing to CD44’s structural and functional diversity. Diseases associated with CD44 dysfunction include superficial keratitis and lichen sclerosus.
