Target Information

LYVE1 has been identified as a major receptor for hyaluronan (HA) on the lymph vessel wall. The deduced amino acid sequence predicts a 322-residue type I integral membrane polypeptide, 41% similar to the CD44 HA receptor. It includes a 212-residue extracellular domain containing a single Link module, the prototypic HA binding domain of the Link protein superfamily.

Like CD44, LYVE1 binds both soluble and immobilized HA. However, unlike CD44, LYVE1 colocalizes with HA on the luminal face of lymph vessels and is absent from blood vessels. Therefore, LYVE1 serves as a lymph-specific HA receptor and a powerful marker for lymphatic vessels.

LYVE-1 is expressed primarily on lymphatic vessel endothelium and may regulate leukocyte and tumor cell trafficking to lymph nodes. The lymphatic vasculature drains extracellular fluid and provides an environment for immune cell responses. Known lymphatic endothelial markers include LYVE1, PALE, VEGFR3, and podoplanin. Diseases associated with LYVE1 dysfunction include Complete Androgen Insensitivity Syndrome.
