Multiplexable

The Click-iT® Plus formulation enhances multiplexing capability compared to the original Click-iT® EdU assays. It can be used with R-PE, R-PE tandems, and fluorescent proteins without compromising accuracy or speed.

Advanced Alternative to BrdU

EdU (5-ethynyl-2´-deoxyuridine) is a thymidine analog incorporated into DNA during synthesis. Detection uses click chemistry—a copper-catalyzed covalent reaction between an azide and an alkyne—providing a more accurate and direct measurement of DNA synthesis than BrdU assays.

Mild Detection Conditions

The small dye-azide molecule enables efficient EdU detection under mild conditions, compatible with standard aldehyde-based fixation and detergent permeabilization. Unlike BrdU assays, DNA denaturation is unnecessary, preserving cell cycle distribution and antigen epitopes. The Click-iT® Plus EdU assay is compatible with cell cycle dyes, antibodies, and standard fluorophores (R-PE, R-PE tandems, GFP, mCherry).

Quick and Simple Protocol

In just five steps, you can prepare samples for flow cytometry analysis:

1. Treat cells with EdU
2. Fix and permeabilize cells
3. Detect S-phase cells using Click-iT® Plus detection cocktail (30 min)
4. Wash once
5. Analyze

Results can be obtained in as little as 60 minutes (recommended: 90 minutes for all applications).
