DNA-directed RNA polymerase II subunit RPB1 (POLR1A)는 DNA 의존성 RNA 중합효소로, DNA를 주형으로 RNA를 합성합니다. POLR1A는 RNA polymerase II의 가장 큰 소단위이며, mRNA 전구체 및 다양한 비암호화 RNA를 합성하는 촉매 구성 요소입니다. 이 효소는 두 번째로 큰 소단위와 함께 활성 중심을 형성하며, 전사 개시 시 DNA 주형 가닥을 중앙 활성 부위로 위치시킵니다. RPB1의 bridging helix는 RNA-DNA 하이브리드를 이동시키는 역할을 하며, 전사 신장 과정에서 Pol II의 이동을 돕습니다. 또한, Pol II의 C-말단 도메인(CTD)의 인산화 상태는 전사 개시, 신장, 종결 및 mRNA 처리 조절에 관여하는 인자들의 결합에 영향을 줍니다.
