Target Information

Tau is a neuronal microtubule-associated protein predominantly located on axons. It promotes tubulin polymerization and stabilizes microtubules. The C-terminus binds axonal microtubules, while the N-terminus interacts with neural plasma membrane components, functioning as a linker between both structures. Tau localization determines axonal polarity in neurons. Short isoforms contribute to cytoskeletal plasticity, while longer ones aid stabilization.

In its hyper-phosphorylated form, Tau becomes the major component of paired helical filaments (PHF), which form neurofibrillary lesions in Alzheimer’s disease (AD) brains. Hyper-phosphorylation disrupts microtubule binding, destabilizing microtubules and leading to neuronal degeneration. Multiple kinases, including GSK-3β, PKA, CDK5, and casein kinase II, phosphorylate Tau. Hyper-phosphorylated Tau is also present in other neurodegenerative disorders such as Pick’s disease, frontotemporal dementia, corticobasal degeneration, and progressive supranuclear palsy.
