• For immunohistochemistry on PFA-fixed tissues, incubate the antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD56 (NCAM, neural cell adhesion molecule) is a transmembrane glycoprotein of the immunoglobulin family that functions as an adhesive molecule. It is widely expressed in the nervous system in isoforms ranging from 120–180 kDa. CD56 is present on T cells and NK cells, contributing to cell migration, axonal growth, pathfinding, and synaptic plasticity.
Polysialic modification reduces CD56-mediated cell adhesion. Through its extracellular region, CD56 mediates homophilic and heterophilic interactions by binding extracellular matrix components such as laminin and integrins.

CD56 is expressed in most neuroectodermal-derived cell lines, tissues, and neoplasms such as retinoblastoma, medulloblastoma, astrocytomas, and neuroblastoma. It is a widely used neuroendocrine marker with high sensitivity for neuroendocrine tumors and ovarian granulosa cell tumors.
Diseases associated with CD56 dysfunction include rabies and blastic plasmacytoid dendritic cell neoplasms.
