c-Abl (Abelson murine leukemia viral oncogene homolog 1, ABL1) is a 140 kDa proto-oncogene tyrosine kinase belonging to the Src family. It plays roles in:

• Cell differentiation, division, adhesion, and stress response
• Cytoskeleton regulation via phosphorylation of CRK, CRKL, and DOK1
• DNA repair and apoptosis regulation under severe DNA damage

The t(9;22) translocation creates a BCR-ABL fusion gene seen in many chronic myelogenous leukemia (CML) cases. ABL1 abnormalities are associated with:

• 90–95% of CML
• 20–25% of adult ALL
• 2–5% of pediatric ALL
