Target Information
GAPDH (Glyceraldehyde-3-phosphate dehydrogenase)는 해당과정(glycolysis)에 관여하는 촉매 효소로, 37 kDa의 동일한 서브유닛 4개로 구성된 테트라머 형태로 존재합니다. NADPH 존재 하에 1,3-bisphosphoglycerate를 glyceraldehyde 3-phosphophate로 가역적으로 환원시키는 반응을 촉매합니다.
GAPDH는 대부분의 조직에서 높게 발현되어 있으며, DNA 복제, DNA 수리, 핵 RNA 수출, 막 융합, 미세소관 결합 등 다양한 세포 내 기능에도 관여합니다. 또한, 세포자멸사(apoptosis), 노화 관련 신경퇴행성 질환, 암 등에서 유전자 발현 조절 및 세포 표현형 형성에 중요한 역할을 하는 것으로 알려져 있습니다.
GAPDH는 알츠하이머병(AD)의 원인 단백질인 amyloid precursor protein 및 헌팅턴병 관련 단백질 Huntingtin과 결합하는 것으로 보고되었으며, actin 및 tubulin과의 상호작용도 알려져 있습니다. 높은 발현 수준으로 인해 Western blot 분석 시 단백질 로딩 컨트롤로 유용하게 사용됩니다.
