Target Information
OTUD7B is a negative regulator of the non-canonical NF-κB pathway that acts by mediating deubiquitination of TRAF3, an inhibitor of the NF-κB pathway. It prevents TRAF3 proteolysis and over-activation of the pathway, thus modulating B-cell responses and mucosal immunity. OTUD7B also deubiquitinates ZAP70, regulating T cell receptor (TCR) signaling and T cell homeostasis. Additionally, it mediates deubiquitination of EGFR and shows activity toward Lys-11, Lys-48, and Lys-63 linked polyubiquitin chains. The physiological significance of its higher catalytic rate with Lys-11-linked chains remains uncertain.
