Target Information

HLA-DR, a major histocompatibility complex (MHC) class II molecule, is a transmembrane glycoprotein composed of a 36 kDa alpha chain (DRA) and a 27 kDa beta chain (DRB).
The alpha chain gene includes five exons:

• Exon 1 encodes the leader peptide
• Exons 2 and 3 encode extracellular domains
• Exon 4 encodes the transmembrane domain and cytoplasmic tail

DRA acts as the sole alpha chain for DRB1, DRB3, DRB4, and DRB5, with polymorphisms found in the beta chain determining peptide-binding specificity.
HLA-DR is mainly expressed on antigen-presenting cells such as B lymphocytes, monocytes, macrophages, thymic epithelial cells, and activated T lymphocytes.
The DR, DQ, and DP loci encode the major human class II region products, which bind processed peptides and present them to T-helper cells, initiating immune responses.

HLA and MHC antibodies are essential tools in Immunopeptidomics, aiding in the identification and characterization of neoantigens using high-performance liquid chromatography coupled with tandem mass spectrometry.
