Target Information

Several serine/threonine protein kinases act as intermediates in signal transduction pathways, including ERK/MAP kinases, ribosomal S6 kinase (Rsk), and Raf-1. Raf-1 is a cytoplasmic serine/threonine kinase broadly expressed in most cell lines and is homologous to v-Raf, the transforming gene product of murine sarcoma virus strain 3611. The unregulated kinase activity of v-Raf is linked to transformation and mitogenesis, while Raf-1 activity is normally suppressed by its N-terminal regulatory domain.
A-Raf, another member of the Raf family, shares substantial homology with Raf-1 within the kinase domain but less elsewhere. A-Raf expression is highest in urogenital tissues and kidney, and lowest in brain tissue.
