Target Information

The accumulation of unfolded proteins in the endoplasmic reticulum (ER) triggers the ER stress response. This response includes inhibition of translation to prevent further accumulation of unfolded proteins, increased expression of proteins involved in the unfolded protein response (UPR), and degradation of misfolded proteins via the ER-associated degradation (ERAD) system.
HERPUD1 may play a role in both UPR and ERAD. Its expression is induced by UPR, and it has an ER stress response element in its promoter region. The encoded protein contains an N-terminal ubiquitin-like domain that may interact with the ERAD system. HERPUD1 interacts with presenilin proteins and can increase amyloid-beta levels upon overexpression. Alternative splicing produces multiple transcript variants, though not all full-length isoforms have been characterized.
