Target Information
Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, upon binding to estrogen, undergoes a conformational change allowing dimerization and binding to estrogen response elements (ERE) in DNA. ER can regulate gene transcription positively or negatively through coactivator or corepressor recruitment.
Two forms of estrogen receptor exist — alpha (ESR1) and beta (ESR2) — encoded by separate genes. At least four isoforms of estrogen receptor-alpha are known (66 kDa, 53 kDa, 47 kDa, 35 kDa) due to alternative RNA splicing. Estrogen receptors are widely expressed and essential for sexual development, reproductive function, and bone maintenance. They are implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
