Target Information
SIAH-2 (seven in absentia homolog 2)는 E3 리가아제로서 단백질 기질의 유비퀴틴화 및 프로테아좀 매개 분해를 촉진합니다. 인간 SIAH-2는 324개의 아미노산으로 구성되며, SIAH-1과 77%, Drosophila sina 유전자와 68%의 서열 동일성을 가집니다. SIAH-2는 TRAF2 및 HIF-1 alpha prolyl hydroxylase 3 (PHD3)를 저산소 조건에서 분해 대상으로 하며, 스트레스 반응 및 사이토카인 신호 조절에 관여합니다. 또한 TNF-alpha 의존적 JNK 활성 및 NFkB 전사 활성화를 감소시킬 수 있습니다. SIAH-2 결핍 생쥐는 저산소 상태에서 호흡 반응 장애 및 헤모글로빈 감소를 보입니다.
