The Pierce™ 660nm Protein Assay Reagent is a ready-to-use, detergent- and reducing agent-compatible reagent for rapid total protein quantitation. It provides a more linear response than Coomassie-based Bradford assays and tolerates higher concentrations of detergents, reducing agents, and other common reagents.

The accessory Ionic Detergent Compatibility Reagent (IDCR) extends detergent compatibility, enabling use with samples containing Laemmli SDS sample buffer and bromophenol blue. Although the Pierce 660-nm Protein Assay shows greater protein-to-protein variation (approximately 37%) compared to the BCA assay, its single-reagent format and broad compatibility make it highly convenient for routine applications.

작동 원리

The assay detects proteins via binding of a proprietary dye-metal complex under acidic conditions, causing a shift in the dye’s absorption maximum to 660 nm. The reddish-brown dye turns green upon protein binding due to deprotonation facilitated by positively charged amino acid groups (histidine, arginine, lysine, and others). The color intensity increases proportionally with protein concentration and remains stable even in the presence of detergents and reducing agents.
