Target Information

The novel protein p53AIP1 (p53-regulated apoptosis-inducing protein 1) has been identified from direct cloning of p53 binding sequences from human genomic DNA. The expression of p53AIP1 in mitochondria is induced by wild-type p53, leading to apoptotic cell death through dissipation of mitochondrial potential. Severe DNA damage causes phosphorylation of p53 at Ser-46, p53AIP1 expression, and apoptotic cell death. DNA damage-inducible apoptosis is enhanced through transcriptional activation of p53AIP1.

P53R2 is directly regulated by p53 to supply nucleotides for DNA repair, playing a key role in cell survival. Dysfunction of this pathway may activate p53-dependent apoptosis to eliminate damaged cells. Conversely, p53AIP1 plays an important role in mediating p53-dependent apoptosis, with phosphorylation at Ser46 regulating transcriptional activation of this apoptosis-inducing gene.
