Target Information
LCK는 Src 계열 티로신 키나아제(Src-family tyrosine kinase)로, T세포 수용체(TCR) 신호전달에 필수적인 역할을 합니다. TCR 자극 시 LCK는 ζ 서브유닛의 ITAM 모티프를 인산화하여 ZAP70의 SH2 도메인 결합 부위를 형성하고, ZAP70을 활성화시켜 LAT 어댑터 단백질을 통한 후속 신호 플랫폼을 생성합니다. 대부분의 LCK는 세포막에 존재하지만, 일부는 골지체에 위치하여 약한 TCR 자극 시 Raf 활성화에 관여할 수 있습니다. LCK는 N-말단의 myristylation 및 palmitylation 부위, PTK 도메인, SH2 및 SH3 도메인을 포함하며, 이는 각각 phosphotyrosine 및 proline-rich 모티프와의 단백질 상호작용을 매개합니다. 또한, LCK는 다양한 자극에 의해 유도되는 세포사멸 조절에도 관여하지만, death receptor에 의한 경로에는 관여하지 않습니다. LCK 관련 질환으로는 Immunodeficiency 22 및 CD45 결핍증이 있으며, 다양한 isoform을 인코딩하는 대체 스플라이싱 변이체가 보고되어 있습니다.
