Product Specific Information
The monoclonal antibody 11A10-B7 recognizes mouse CD301b (macrophage galactose-type C-type lectin 2, MGL2). CD301b expression is restricted to dermal dendritic cells, connective tissue, and bone marrow-derived dendritic cells. Macrophages also express CD301b at low levels.
CD301b contains a carbohydrate recognition domain that binds protein antigens with galactose and N-acetylgalactosamine (Gal/GalNAc). These antigens are internalized via CD301b and presented to CD4+ T cells through MHC Class II. CD301b+ dendritic cells play an important role in TH2 priming.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse skin draining lymph node cells at ≤ 0.25 µg/test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Optimize titration for best performance.
Fluorochrome Information:
eFluor® 660 is a replacement for Alexa Fluor® 647. Emits at 659 nm and is excited by the red laser (633 nm). Ensure instrument compatibility.
Excitation: 633–647 nm; Emission: 668 nm; Laser: Red.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
CD301b (MGL2) is expressed mainly in dermal and bone marrow-derived dendritic cells. It mediates internalization of Gal/GalNAc-containing antigens and contributes to TH2 immune response priming.
