| Cellular Localization | Cytoplasm, Nucleus. Shuttles between cytoplasm and nucleus. Largely nuclear in unstimulated cells. In osteoblasts, colocalizes with ATF4 and RUNX2 in the nucleus. Serum deprivation increases nuclear localization, activating SOX9 expression and chondrogenesis. Insulin-induced phosphorylation at Ser-256 by PKB/AKT1 leads to nuclear export and degradation. Phosphorylation at Ser-249 by CDK1 promotes nuclear accumulation. |
| Background | Chromosomal aberrations involving FOXO1 are a cause of rhabdomyosarcoma 2 (RMS2), also known as alveolar rhabdomyosarcoma. Translocation (2;13)(q35;q14) with PAX3 or t(1;13)(p36;q14) with PAX7 results in transcriptional activator proteins. FOXO1 functions as a transcription factor. Phosphorylated by AKT1; insulin-induced. IGF1 induces phosphorylation at Ser-256, Thr-24, and Ser-319, decreasing DNA-binding activity and promoting nuclear exclusion. Phosphorylated upon DNA damage, likely by ATM or ATR. Contains one fork-head DNA-binding domain. Interacts with LRPPRC. Ubiquitously expressed. |
