Cellular Localization Isoform 1: Nucleus, Cytoplasm, Cell membrane (Peripheral membrane protein, Cytoplasmic side). A minor fraction is associated with the inner membrane.
Isoform 3: Nucleus, Cytoplasm, Cell membrane (Peripheral membrane protein, Cytoplasmic side). Single-pass type I membrane protein associated with inner membrane via palmitoylation (Probable). Subset exists as transmembrane protein with N-terminal extracellular domain.
Also localized in Nucleus, Golgi apparatus, and plasma membrane when palmitoylated.

Background

  • Domain: Composed of three domains: N-terminal modulating domain, DNA-binding domain, and C-terminal steroid-binding domain.
  • Function: Nuclear hormone receptor regulating eukaryotic gene expression, affecting cellular proliferation and differentiation in target tissues.
  • Polymorphism: Genetic variations in ESR1 correlate with bone mineral density (BMD). Low BMD increases risk of osteoporotic fracture.
  • PTM: Glycosylated (contains N-acetylglucosamine, probably O-linked); phosphorylated by cyclin A/CDK2 to enhance transcriptional activity.
  • Similarity: Member of the nuclear hormone receptor family (NR3 subfamily); contains one nuclear receptor DNA-binding domain.
  • Subunit: Interacts with multiple proteins including SLC30A9, ESR2, NCOA3/5/6, NCOA7, PHB2, PELP1, UBE1C, AKAP13, CUEDC2, KDM5A, SMARD1, HEXIM1, MAP1S, PBXIP1, MUC1 (estradiol-stimulated), DNTTIP2, FAM120B, UIMC1, TXNRD1 isoform 4, MLL2, and ATAD2 (estradiol-enhanced interaction).
