배경 (Background)
- Catalytic activity: Cleaves the triple helix of collagen at approximately three-quarters of its length from the N-terminus (775-Gly-|Ile-776 in alpha-1(I) chain).
- Substrate specificity: Cleaves collagens of types I, II, III, VII, and X.
- Cofactors: Binds 2 zinc ions and 4 calcium ions per subunit.
- Domain structure: Contains catalytic N-terminal and C-terminal domains; C-terminal domain binds TIMP (tissue inhibitor of metalloproteinases).
- Regulation: Can be activated without removal of the activation peptide.
- Function: Involved in extracellular matrix remodeling and collagen degradation; interacts with HIV-1 Tat protein, reducing Tat-mediated neurotoxicity.
- Post-translational modification (PTM): Undergoes autolytic cleavage to major forms (22 kDa and 27 kDa); minor glycosylated form (25 kDa) derived from 22 kDa form.
- Similarity: Belongs to the peptidase M10A family; contains 4 hemopexin-like domains.
