Target Information
The 6x His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH). It is commonly expressed at either the N- or C-terminal regions of recombinant proteins to allow isolation or purification by immobilized metal affinity chromatography (IMAC).
Epitope tagging enables detection of recombinant proteins lacking specific antibodies by fusing a known epitope to the protein. His-tagged proteins are purified using metal chelate chromatography due to the metal-binding properties of histidine clusters. The affinity of the His-tag motif to Ni²⁺ allows efficient purification on Ni²⁺-NTA adsorbents. His tag-specific antibodies facilitate detection and co-immunoprecipitation of tagged proteins.
