MAGEB1 유전자는 MAGEB 유전자 패밀리에 속하며, 이 패밀리의 구성원들은 전체 코딩 서열이 마지막 엑손에 위치하고 있습니다. 이 단백질들은 상호 간 50~68%의 서열 유사성을 보이며, 프로모터와 첫 번째 엑손의 다양성으로 인해 동일한 기능이 서로 다른 전사 조절 하에서 발현될 수 있습니다.
이 유전자는 DSS(dosage-sensitive sex reversal) 영역에 위치하며, 고환과 다양한 종양에서 발현됩니다. MAGEB 계열 유전자는 X 염색체 Xp22-p21 영역에 클러스터되어 있습니다. 여러 스플라이싱 변이체가 보고되었으나, 일부의 전체 길이는 아직 정의되지 않았습니다.
