Target Information
GAD-65와 GAD-67은 group II decarboxylase 계열 단백질로, L-glutamic acid로부터 GABA (gamma-aminobutyric acid)를 생성하는 속도 제한 단계를 촉매한다.
두 GAD 단백질 모두 뇌에서 발견되며, GAD-65는 신경 말단의 시냅스 소포막에, GAD-67은 세포 전체에 분포한다.
GAD-67은 기본적인 GABA 합성을 담당하며, 신경전달 시 수요가 증가할 경우 GAD-65가 일시적으로 활성화되어 GABA 생산을 보조한다.
GAD-65의 결손은 GABA 신경전달을 저해할 수 있으며, GAD-67의 결손은 치명적이다.
대체 스플라이싱으로 인해 GAD-67의 두 가지 아이소폼(GAD-67, GAD-25)이 존재한다.
