Target Information
The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. PR mediates the physiological effects of progesterone, which plays a central role in reproductive events associated with the establishment and maintenance of pregnancy.
PR exists as two forms:
- PR-A: 94 kDa protein with a 165 amino acid deletion at the N-terminus, containing an inhibitory domain that represses steroid hormone transcriptional activity.
- PR-B: 120 kDa protein, transcriptionally active form responsible for activating genes related to endometrial maintenance, pregnancy, and inhibition of ovulation.
In its inactive state, PgR forms a multiprotein complex with heat shock proteins and immunophilins. Upon progesterone binding, conformational changes lead to dimerization and binding to progesterone response elements (PRE), activating transcription. Null mutations in the PGR gene can result in reproductive abnormalities.
