This product is diluted and ready-to-use.
A recommended positive control tissue is Rhabdomyosarcoma, but other positive controls may also be suitable.

The primary antibody is intended for professional laboratory use for detecting the corresponding protein in formalin-fixed, paraffin-embedded tissues using manual qualitative immunohistochemistry (IHC). It is used after primary tumor diagnosis by conventional histopathology.

MyoD1 Function and Diagnostic Utility
MyoD1, a member of the myogenic helix-loop-helix transcription factor family, coordinates myogenic differentiation—from mesodermal precursors to myoblasts, myotubes, and mature skeletal myofibers.
Normal mature skeletal muscle does not express MyoD1 protein. MyoD1 is expressed in myoblasts before differentiation, while myogenin functions post-differentiation.
Anti-MyoD1 immunostaining identifies early myogenic cells and serves as a biomarker for less differentiated Rhabdomyosarcomas (RMS). RMS are common malignant soft tissue neoplasms in childhood.
For diagnostic accuracy, anti-MyoD1 may be used in combination with anti-Myogenin and anti-Desmin as a panel of markers, since RMS rarely shows negativity for all three simultaneously.

IHC Procedure Notes
Use on formalin-fixed, paraffin-embedded sections. Pretreatment with heat-induced or enzymatic epitope retrieval is recommended.
IHC staining visualizes antigens via sequential application of primary antibody, secondary antibody, enzyme complex, and chromogenic substrate. The visible reaction product is observed under a light microscope to aid differential diagnosis.

Control Recommendations

  • A positive tissue control should be included in every staining procedure.
  • Controls should include both positive and negative staining areas.
  • External positive controls should be freshly prepared and processed similarly to patient samples.
  • Weak positive tissues are ideal for detecting subtle changes in antibody sensitivity or reagent degradation.
  • Internal or external negative controls should verify absence of specific staining and help assess background staining.

Target Information

The MyoD gene was first identified for its ability to convert embryonic mouse fibroblasts into muscle cells. Forced expression of MyoD (human homolog myf3) activates muscle-specific genes even in non-muscle cells.
Its regulatory domain includes a 70 amino acid region containing a basic DNA-binding motif and a helix-loop-helix (HLH) dimerization motif.
Other genes with homologous HLH domains include myf5, myogenin (myf4), and myf6 (MRF4/herculin).
