Target Information
Thymidylate Synthase (TS) catalyzes the conversion of deoxyuridine monophosphate (dUMP) to deoxythymidine monophosphate (dTMP), essential for DNA biosynthesis. TS is a critical target for fluoropyrimidines, a group of antineoplastic drugs used in treating solid tumors. 5-FU and fluorodeoxyuridine are converted in tumor cells to FdUMP, which inactivates TS via a ternary covalent complex with 5,10-methylenetetrahydrofolate. TS expression correlates with response to 5-fluorouracil (5-FU) in various human carcinomas (colorectal, gastric, head and neck, breast).
