Target Information
RMP (RNA polymerase II, subunit 5-mediating protein) is involved in gene transcription regulation. It acts as a transcriptional repressor with the corepressor UXT to regulate androgen receptor (AR) transcription and may function as a tumor suppressor by repressing AR-mediated gene transcription and inhibiting anchorage-independent growth in prostate cancer cells. It is required for cell survival in ovarian cancer cells and, together with UXT, associates with chromatin at the NKX3-1 promoter region. RMP antagonizes transcriptional modulation via hepatitis B virus X protein and plays a central role in maintaining S6K1 signaling and BAD phosphorylation under normal growth conditions, thereby protecting cells from deleterious effects of sustained S6K1 signaling. The URI1-PPP1CC complex serves as a central component of a negative feedback mechanism counteracting excessive S6K1 survival signaling to BAD in response to growth factors. It mediates inhibition of PPP1CC phosphatase activity in mitochondria and coordinates nutrient-sensitive gene expression in a mTOR-dependent manner. RMP appears to act as a scaffolding protein assembling a prefoldin-like complex containing PFDs and proteins involved in transcription and ubiquitination. [Uniprot]
