Parkinson’s disease is the second most common neurodegenerative disorder after Alzheimer’s. It affects approximately 1% of people over 65 and 3% over 75. Mutations in the Park2 gene are a major cause of Parkinson’s disease. Although its function is not fully understood, Park2 may play a role in the ubiquitin-mediated proteolytic pathway. Mutations are associated with autosomal recessive juvenile parkinsonism. Alternative splicing of Park2 results in three known products of undetermined function. Panneuronal expression of Parkin substrate Pael-R leads to age-dependent degeneration of dopaminergic neurons in Drosophila; coexpression with Parkin degrades Pael-R and suppresses its toxicity.
