Background
This gene encodes a cysteine protease with strict specificity for hydrolysis of asparaginyl bonds. The enzyme is involved in processing bacterial peptides and endogenous proteins for MHC class II presentation in lysosomal/endosomal systems. Activation occurs under acidic pH and is autocatalytic. Expression is induced when monocytes differentiate into dendritic cells, and a fully active enzyme is produced following lipopolysaccharide stimulation. Overexpression is associated with many solid tumor types. A pseudogene exists on chromosome 13, and multiple alternatively spliced variants have been reported, with two validated isoforms encoding the same protein.
