Background
The RYK gene encodes an atypical member of the growth factor receptor protein tyrosine kinase family, differing from other members in conserved residues of activation and nucleotide binding domains. It belongs to a subfamily not regulated by phosphorylation in the activation segment. Biological activity may involve recruitment of a signaling-competent auxiliary protein via an uncharacterized mechanism. The encoded protein features a leucine-rich extracellular domain with a WIF-type Wnt binding region, a single transmembrane domain, and an intracellular tyrosine kinase domain. RYK is implicated in Wnt signaling pathways, including axon pathfinding regulation. Alternative splicing generates multiple transcript variants encoding distinct isoforms.
