Background
Defects in CFC1 are associated with several congenital heart malformations (CTHM) including tetralogy of Fallot, pulmonary atresia, double-outlet right ventricle, truncus arteriosus communis, and aortic arch anomalies.
Mutations in CFC1 also cause transposition of the great arteries (DTGA), leading to complete inversion of the great vessels and incompatible circulatory separation, often requiring surgical intervention.
Additionally, CFC1 defects are linked to visceral heterotaxy (HTX2), characterized by complex cardiac malformations and situs inversus or situs ambiguus.
Functionally, CFC1 is involved in establishing the left-right axis and may play a role in mesoderm and neural patterning during gastrulation.
Post-translational modification includes N-glycosylation, and the protein contains one EGF-like domain.
