Background

  • Alternative products: Additional isoforms seem to exist.
  • Domain: Two independent domains — LEM-like (LAP2-N or LEM-D) and LEM (LAP2-C or LEM-B). LEM-like binds DNA, LEM interacts with BANF1.
  • Function: Involved in structural organization of the nucleus and post-mitotic nuclear assembly. Plays an important role with LMNA in nuclear anchorage of RB1.
  • Role in nuclear lamina: Helps direct assembly and maintain structural organization of the nuclear envelope. Possible receptor for lamin filament attachment.
  • DNA replication: May regulate initiation through interaction with NAKAP95.
  • Pharmaceutical relevance: TP5 available as Timunox, Sintomodulina, and Mepentil — used for immune deficiencies, autoimmunity, infections, and cancer.
  • PTM: Mitosis-specific phosphorylation abolishes binding to lamin B and chromosomes; phosphorylated in a mitosis-specific manner.
  • Similarity: Member of LEM family; contains 1 LEM domain and 1 LEM-like domain.
  • Subcellular location: Diffuse nuclear expression; tightly associated with nuclear lamina.
  • Subunit interactions: Interacts with LMNA, LMNB1, LMNB2, BANF1, RB1, NAKAP95, GMCL, and chromosomes.
  • Tissue specificity: Expressed in many tissues; most abundant in adult thymus and fetal liver.
