Target Information
Focal Adhesion Kinase (FAK) is a 125 kDa non-receptor protein tyrosine kinase that functions as a substrate for Src and plays a key role in integrin-mediated signaling. It regulates cell spreading, differentiation, migration, apoptosis, and cell cycle progression. FAK localizes to focal adhesions, which are sites of cell-extracellular matrix interactions via integrin receptors. Upon integrin engagement with fibronectin, FAK undergoes increased tyrosine phosphorylation, promoting interactions with Grb2 and c-Src, leading to ERK2 MAP kinase activation. Multiple transcript variants encoding different isoforms of FAK have been identified.
