Target Information
Tryptases comprise a family of trypsin-like serine proteases, the peptidase family S1. These enzymes are active only as heparin-stabilized tetramers and resistant to endogenous proteinase inhibitors. Multiple tryptase genes are clustered on chromosome 16p13.3, characterized by conserved 3' UTRs and tandem repeats that may regulate mRNA stability. Most tryptase genes contain an intron upstream of the initiator Met codon, separating transcription initiation from coding sequence—a feature typical of tryptases but rare in other genes. Tryptases are implicated in asthma and other allergic or inflammatory disorders. TPSD1, previously considered a pseudogene, is now thought to encode a functional protein.
