Casein kinase I (CKI) and casein kinase II (CKII) are families of serine/threonine protein kinases found in all eukaryotic organisms. Members of the CKI family, including CKI alpha, gamma, epsilon, and delta, are involved in regulating cytoplasmic and nuclear processes such as DNA replication and repair.
CKII typically exists as a tetrameric complex (α2β2 or αα′β2), where the catalytic α subunit is regulated by the β subunit, which can autophosphorylate. CKII shows high activity in proliferating and differentiating cells, phosphorylating over 100 substrates including nuclear oncoproteins, transcription factors, and DNA metabolism enzymes.
