Product Specific Information

The 53-6.7 monoclonal antibody reacts with the mouse CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8αβ heterodimer or CD8αα homodimer.
CD8 binds MHC class I and associates with protein tyrosine kinase p56lck, playing a role in T cell development and activation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes (≤ 0.5 µg/test)

Usage Recommendation:

• Titrate antibody for optimal performance.
• Define cell number empirically (10⁵–10⁸ cells/test).
• Use CellBlox Plus Blocking Buffer for optimal staining (5 µL/sample).

Compatibility Notes:

• NovaFluor dyes are not compatible with DNA intercalating dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

Strain-specific observation:
NovaFluor Yellow 660 may non-specifically stain B cells in SJL mice; not observed in BALB/c or C57BL/6.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein involved in T cell differentiation and activation.
It functions as a co-receptor with TCR for MHC class I molecules and is expressed on T cells, NK cells, monocytes, and dendritic cells.
CD8α chain is essential for MHC-I binding, and activation leads to CTL differentiation and pathogen/tumor clearance.
CTL differentiation is enhanced by IL-2, IL-12, and TGF-β1.
