Target Information

Janus kinase 2 (JAK2) is a protein tyrosine kinase involved in specific cytokine receptor signaling pathways and is known to be mutated in polycythemia vera. This kinase contains the catalytic domain (JH1) of JAK2. Activation of JAKs leads to phosphorylation of STAT (signal transducers and activators of transcription) proteins, which dimerize and translocate to the nucleus to modulate gene transcription, including interferon-stimulated genes.
JAK2 is required for IFN gamma-receptor complex initiation, while JAK1 acts as an amplifier. Some studies suggest that Tyk2 and JAK3 could substitute for JAK2 if positioned correctly within the receptor complex. Mutations in JAK2 are associated with Budd-Chiari syndrome, Polycythemia vera, Thrombocythemia 3, myelofibrosis, and Acute myelogenous leukemia.
