Target Information
SLP-76 is a substrate of the ZAP-70 protein tyrosine kinase activated upon T cell receptor (TCR) ligation. The SLP-76 gene is located on human chromosome 5q33 and encodes a 533 amino acid adaptor protein comprising:
- An N-terminal acidic region with a PEST domain and multiple tyrosine phosphorylation sites
- A central proline-rich domain
- A C-terminal SH2 domain
SLP-76 interacts with several proteins constitutively and upon receptor activation, functioning as a scaffold in T cell signaling. Studies using SLP-76 deficient models indicate its essential role in T cell development, activation, and mast cell/platelet function.
