Target Information

The human UNG gene encodes both nuclear (UNG2) and mitochondrial (UNG1) forms of uracil-DNA glycosylase. These isoforms are produced by alternative splicing and use of two differentially regulated promoters (PA and PB).
UNG1 and UNG2 cDNAs are similar in size (2061 and 2058 bp, respectively), and thus their mRNAs are not resolved as separate species in gel electrophoresis.
Nuclear UNG2 differs from mitochondrial UNG1 by 44 amino acids at the N-terminal region, which are not required for catalytic activity.

A major role of UNG gene products is to repair mutagenic U:G mispairs caused by cytosine deamination. For example, UNG2 removes misincorporated dUMP residues.
The expression levels of UNG1 and UNG2 vary between cell and tissue types and are regulated by the cell cycle. Generally, UNG expression and activity are higher in proliferating cells compared to non-proliferating ones.
UNG1 is a 304 amino acid protein, while UNG2 consists of 313 amino acids.
