Target Information
Protein p21 is a tumor suppressor protein that inhibits Cyclin-dependent kinases (CDK) and is transcriptionally activated by p53.
It plays a critical role in the cellular response to genotoxic stress and serves as a major transcriptional target of p53.
In the nucleus, p21 binds and inhibits Cdk1 and Cdk2, blocking cell cycle progression from G1 to S phase or G2 to mitosis after DNA damage, allowing DNA repair.
In the cytoplasm, p21 exhibits anti-apoptotic effects by inhibiting caspase 3 and apoptotic kinases ASK1 and JNK.
p21 has dual roles in carcinogenesis — functioning both as a tumor suppressor and as an oncogene depending on cellular context.
