Target Information

Proposed scaffold protein implicated in diverse physiological processes. COMMD1 regulates ubiquitination of specific cellular proteins and modulates the activity of cullin-RING E3 ubiquitin ligase (CRL) complexes by displacing CAND1. It promotes ubiquitination and proteasomal degradation of NF-κB subunit RELA, downregulating NF-κB activity. COMMD1 is involved in membrane expression and ubiquitination of SLC12A2 and regulates Na⁺ transport in epithelial cells through modulation of ENaC subunit expression and ubiquitination via NEDD4L. It promotes localization of SCNN1D to recycling endosomes and enhances CFTR cell surface expression through ubiquitination regulation. COMMD1 also downregulates SOD1 activity by interfering with homodimerization and plays a role in copper ion homeostasis, including copper-dependent ATP7A trafficking. It binds one copper ion per monomer and may facilitate biliary copper excretion within hepatocytes. Additionally, COMMD1 binds phosphatidylinositol 4,5-bisphosphate (PtdIns(4,5)P2) and regulates HIF1A-mediated transcription by competing with ARNT/HIF-1β for HIF1A binding, resulting in decreased DNA binding and impaired transcriptional activation by HIF-1.
