The Spike(S) protein is critical for viral attachment, fusion, and entry, making it a key target for antibody and vaccine development.
The receptor-binding domain (RBD, S-RBD) in the Spike protein binds strongly to ACE2 receptors in humans and bats, exhibiting higher affinity compared to SARS-CoV RBD.
S1 subunit contains the RBD that specifically binds to ACE2, while S2 mediates membrane fusion.
SARS-CoV-2 Spike protein is 1273 amino acids long, consisting of:

• Signal peptide (1–13)
• S1 subunit (14–685)
• S2 subunit (686–1273)

The S-ECD refers to the extracellular domain of the Spike protein.
