A recommended positive control tissue for this product is Colon adenocarcinoma (Lynch/MLH-1); however, other positive controls may be used.
This antibody is intended for laboratory professional use in detecting MLH1 protein in formalin-fixed, paraffin-embedded tissues via manual qualitative IHC testing. It should be applied after primary tumor diagnosis by standard histopathology.
This monoclonal antibody recognizes an 83 kDa MLH1 protein, a key component of the DNA mismatch repair system (MMR). MLH1 defects are associated with hereditary non-polyposis colorectal cancer type 2 (HNPCC2). MLH1 forms heterodimers with PMS2 (MutL alpha) and MLH3 (MutL gamma), playing roles in DNA repair and meiosis.
Pretreatment of deparaffinized tissue with heat-induced epitope retrieval or enzymatic retrieval is recommended. IHC staining visualizes antigens through sequential antibody binding, enzyme complex formation, and chromogenic substrate development. Results should be interpreted microscopically to aid in differential diagnosis.
A positive tissue control must be included in each staining procedure. Tissues with weak positive staining are best suited for detecting subtle changes in antibody sensitivity or staining quality.
Negative control tissues (internal or external) should confirm absence of specific staining and help identify non-specific background staining. If specific staining appears in negative controls, patient specimen results are invalid.

MLH1 is a DNA mismatch repair protein essential for maintaining genetic stability. Defects cause microsatellite instability (MSI) and are linked to carcinogenesis, including hereditary nonpolyposis colon cancer (HNPC). MLH1 works with MSH2 and PMS2 in the repair process, ensuring fidelity during DNA replication.
